HIGHLIGHT OF BUFFALO MILK CHOLESTEROL THROUGH THIN-LAYER CHROMATOGRAPHY

Abstract
The highlight of buffalo milk cholesterol was conducted by means of thin-layer chromatography and the migration of mobile phases along an absorbent layer. Free and esterised cholesterol extraction was conducted according to the method of Folch et. al., performing small alterations regarding solvent volume or the volume of samples under analysis. Many reactives underwent testing, as the initial iodine visualising was not appropriate. The quantitative identification was conducted by comparing the Rfs of free and esterised cholesterol from analysed samples with existing standards. Spots were highlighted using a photodensimeter.

EVIDENTIEREA COLESTEROLULUI DIN LAPTELE DE BIVOLITA PRIN CROMATOGRAFIE ĪN STRAT SUBTIRE

Rezumat
Evidentierea colesterolului din laptele de bivolita s-a realizat prin cromatografie īn strat subtire prin migrarea unei faze mobile de-a lungul unui strat absorbant. Extractia colesterolului liber si esterificat din laptele de bivolita s-a efectuat dupa metoda lui Folch si col., cu modificari minore privind volumele solventilor sau a probelor analizate. Au fost testati mai multi reactivi deoarece vizualizarea intiala cu iod nu a fost corespunzatoare. Identificarea calitativa s-a facut prin compararea Rf-urilor colesterolului liber si esterificat din probele analizate cu standardele existente. Spoturile au fost evidentiate cu ajutorul unui fotodensimetru.
